Manipulation of bacteria – Part 2
In the previous practical, students gained experience with transformation of competent E. coli, the streak plate procedure and viable plate count methods. In this practical, students will have the opportunity to record the results from the incubated plates and stain the cellulose plates to detect cellulase activity.
Materials required
• Streak plates & viable cell count plates inoculated at previous practical
• Marker pens to aid colony counting
• 50 to 100 ml congo red (1 mg ml-1 )
• Approximately 200 ml 1M NaCl
• 25 ml pipettes and pipettors.
Prior preparation
Plates inoculated by students in the previous class were incubated overnight at 37°C and then stored at 4°C until required in this class.
